MagNA Pure 96 System
Discover what high throughput is all about
Treat all your samples like urgent
samples
Process 1 to 96 standard volume samples in less
than one hour including the handling time that is
needed for the setup of a run. Larger volumes
require typically up to 80 to 90 minutes for a run.
The MagNA Pure 96 Instrument uses two
pipetting arms, handling all 96 samples absolutely
standardized at the same time.
System
Convenient instrument loading using
racks and automated vortexing
The few barcoded, prefilled ready-to-use reagent
trays and consumables are loaded into racks for
increased ease and user safety. Simply use a
second set of racks to exchange reagent trays and
purify cellular RNA after isolating DNA.
The onboard magnetic glass particle (MGP) shaker
eliminates manual MGP vortexing.
G Figure 1: A reagent rack in the process of being
loaded with reagent trays
Barcode-supported reagent and
consumables verification
Barcode reading is integrated throughout the entire
purification procedure using a barcode scanner,
minimizing the risk of user errors during loading
and enabling full process documentation.
Flexible elution formats
Purified DNA, RNA or viral nucleic acids are eluted
into the actively cooled MagNA Pure 96 Output
Plate. A passively cooled custom plate can also be
selected for the 96-format, e.g., for archiving or
further downstream use.

Intelligent features
Networked data workflow
Automatic rollover of reagents, by which a second
set of reagent trays is automatically used when the
first set is used up in the same run, is an integral
part of the MagNA Pure 96 System. Automated load
check saves time by detecting an error and checking
only the rack(s) that had been corrected by the user.

Exchange data bidirectionally with the Laboratory
Information Management System (LIMS) and
share files in *.xls format.
Entire run data files, or selected run data are
configurable outside the MagNA Pure 96
Software environment for loading before the
run is started.
Export selective or complete run-results for easy
post-purification run documentation or transfer
to another instrument in the workflow.
Consistent data workflow from sample to result
maintains data integrity, avoiding redundant
data handling.
Software
Optimized application protocols
Preprogrammed purification protocols with flexible
parameter options eliminate the time-consuming
steps typically required to program a fully optimized
application.
Full traceability and easy data backups
All barcoded parameters are fully traceable with
an available audit track and CFR21.11 compliant.
Take advantage of seamless experimental data
workflow in combination with Roche Instrument
Systems, such as the LightCycler® 480 RealTime PCR System, featuring direct sample name
uploading to LightCycler® Software.

Experience how easy MagNA Pure 96 Kits are to
use. Simply position the magnetic glass particles
(MGP) and trays prefilled with reagents on the
instrument stage*. MGP vortexing is automated by
the MagNA Pure 96 System’s integrated shaker.
Purification run setup is possible in less than
5 minutes. Reagent trays are automatically pierced by
metal needles of the MagNA Pure 96 Instrument’s
robotic dispensing head.
Sample numbers are selectable from 1 to 96.
Reagent tray reuse is very convenient. Evaporationproof sealing foils are easily applied, and MGP
bottles are self sealing.
Sealed reagent trays can be stored at room temperature (+15 to +25°C) for 2 to 4 weeks, depending on
kit type, for later use. Opened reagent trays remain
stable unsealed on the instrument overnight.

MagNA Pure 96 DNA and Viral NA Large Volume Kit
Isolate high quality genomic DNA or
viral DNA and RNA for PCR or Real-time
PCR of sample volume up to 1000 µl
This kit is recommended for highly sensitive
applications where large sample volumes are used.
Typical yields for genomic DNA purification
from whole blood are 11–17 µg, and 22 µg from
culture cells (106 K-562 cells).

Prevention of Cross Contamination
In this experiment, 96 plasma samples of
500 µl sample volume were processed on the
MagNA Pure 96 Instrument in an alternating
(checkerboard) pattern in which samples in
one well were spiked with a high concentration of
Parvo B19 plasmid, and the neighboring wells were
not spiked. All isolates were analyzed for the presence
of Parvovirus B19 DNA using the LightCycler® 480
Instrument and a 384-well PCR plate:
